No detectable DNA, RNA and protein traces.<\/p>\n<p><b>Purification Method<\/b><\/p>\n<p><b>S-form LPS <\/b>S-form LPS was isolated by the hot phenol-water method followed by a modified phenol-chloroform-petroleumether method. Semi-purified LPS was subjected to further re-extraction cycles and ultracentrifugation steps, extensively electrodialysed before converted to its uniform sodium salt form to yield TLRpure\u2122 LPS.<\/p>\n<p><b>R-form LPS <\/b>R-form (mutant\/rough) LPS was isolated by phenol-chloroform-petroleum-ether method. Semi-purified LPS was subjected to further re-extraction cycles and ultracentrifugation steps, extensively electrodialysed before converted to its uniform sodium salt form to yield TLRpure\u2122 LPS.<\/p>\n<p><b>MPLA <\/b>MPLA was generated by acid hydrolysis from R-form TLRpure\u2122 LPS.<\/p>\n<p><b>Concentration\u00a0\u00a0 <\/b><br \/>\n1mg\/ml (0.5mg\/ml for 250\u03bcg size) stabilised in sterile, double-distilled water (ddWater), without any additives.<\/p>\n<p><b>Biological Activity <\/b><br \/>\nOptimal concentration is dependent upon cell type, species, desired activation and analysis: 0.01-1.0\u03bcg\/ml. Prepare diluted MPLA working solutions in water just prior to use, keep sterile. Do <i>not <\/i>pre-dilute in buffer (e.g. PBS) as this will lead to precipitation of MPLA. To yield a 100\u03bcg\/ml (1,000-100x) stock solution add 100\u03bcl of Lipid A to 900\u03bcl endotoxin-free sterile\u00a0 ddWater (Cat. No.: IAX-900-002) (<i>not <\/i>PBS) and mix well.
